Biological activity MDX-1401 binds to recombinant CD30 with a K d value of 0.83 nmol/L, exhibiting binding affinity comparable to that of its parental antibody MDX-060 [1]. MDX-1401 (125 I-MDX-1401) binds to CD30 on the surface of the Hodgkin's lymphoma cell line L540 cells, with a K d value of approximately 1.9 nmol/L [1]. MDX-1401 (0.012-50 ug/mL; 30 min primary incubation, 30 min secondary incubation) binds to CD30 + lymphoma cell lines (L540, Karpas-299, L428, L1236) in a dose-dependent manner, and its binding properties are consistent with those of the parental antibody MDX-060 [1]. MDX-1401 mediates FcγRIIIa-dependent antibody-dependent cellular cytotoxicity (ADCC) against L540 Hodgkin lymphoma cells, with an EC 50 of 0.009 ug/mL and a maximum cell lysis rate of 87%, exhibiting superior potency and efficacy compared to the parental antibody MDX-060 [1]. MDX-1401 (treated at 37°C for 4 h) mediates dose-dependent ADCC against CD30 + lymphoma cell lines (L540, Karpas-299, L428, L1236), with EC 50 values ranging from 0.008 to 0.04 ug/mL and maximum lysis rates of 13% to 68%. This includes activity against L1236 cells with low CD30 expression, against which the parental antibody MDX-060 shows no activity [1]. MDX-1401 binds to CD30 on L540 Hodgkin lymphoma cells with a K D value of 1.9 nM, and its antigen-binding affinity is nearly identical to that of its parental antibody MDX-060 (K D ~1.4 nM) [2]. MDX-1401 mediates significantly stronger and more effective ADCC against CD30-expressing tumor cell lines (including those with low CD30 expression) across all FcgRIIIa polymorphic variants, with a decreased EC 50 and an increased maximum specific lysis rate compared to its parental antibody [2]. MDX-1401 (1 mg/kg; i.p.; single dose) significantly prolongs the survival of SCID mice with systemic Karpas-299 ALCL [1].
