CAN10 effectively impairs the proinflammatory and profibrotic effects of IL1RAP-related cytokines (IL-1, IL-33, and IL-36) in human umbilical cord endothelial cells and human dermal fibroblasts, which are critical to systemic sclerosis pathophysiology [1]. CAN10 (mCAN10, mouse surrogate of CAN10) (10-20 mg/kg; i.p.; twice weekly; 7 weeks) reduces cutaneous and pulmonary fibrosis in a murine cGvHD model of systemic sclerosis, with significant improvements in dermal thickness, hydroxyproline content, myofibroblast counts, and lung Ashcroft scores [1]. |
