Immunofluorescence
Jurkat cells were fixed with 4% paraformaldehyde for 20 minutes, followed by blocking with 5% goat serum for 1 hour. The cells were then incubated with a primary antibody targeting L-Selectin/CD62L (HB644013) at a concentration of 20 ug/ml overnight at 4°C. Afterward, the cells were incubated with a secondary antibody, Goat Anti-Rabbit IgG (Alexa Fluor-488) at a concentration of 6.5 ug/ml for 50 minutes at room temperature. The localization of L-Selectin/CD62L was visualized (shown in green), and nuclear DNA was stained with DAPI (shown in blue).

Flow cytometry
Flow-cytometry using anti-Human CD62L antibody. Loucy cells were stained with an irrelevant antibody (Blue Histogram) or an anti-Human CD62L monoclonal antibody (Catalog HB644013, Yellow Histogram) at a concentration of 5 µg/ml for 30 mins at RT. After washing, bound antibody was detected using a Goat Anti-Rabbit IgG H&L Polyclonal Antibody, FITC (abinScience:TF690414) and cells analysed on a NovoCyte Flow Cytometer.
