Western blot
Western blot analysis was performed using anti-PD-L1 monoclonal antibody at 1 ug/mL on various samples.
Lane 1: PD-L1-overexpressing HEK293T cell lysate

Immunofluorescence
293T cells overexpressing PD-L1 were fixed with 4% paraformaldehyde for 20 minutes, followed by blocking with 5% goat serum for 1 hour. The cells were then incubated with a primary antibody targeting PD-L1 (HV974133) at a concentration of 20 ug/ml overnight at 4°C. Afterward, the cells were incubated with a secondary antibody, Goat Anti-Rabbit IgG (Alexa Fluor-488) at a concentration of 6.5 ug/ml for 50 minutes at room temperature. The localization of PD-L1 was visualized (shown in green), and nuclear DNA was stained with DAPI (shown in blue).

Flow cytometry
Flow-cytometry using anti-Human PD-L1 antibody. Overexpression cells and Untransfected cells were stained with an anti-Human CD274 monoclonal antibody (Catalog HV974133, Yellow Histogram) at a concentration of 5 µg/ml for 30 mins at RT. After washing, bound antibody was detected using a Goat Anti-Rabbit IgG H&L Polyclonal Antibody, FITC (abinScience:TF690414) and cells analysed on a NovoCyte Flow Cytometer.
