Adjuvants shape the quality and durability of vaccine responses through early innate immune programs. Using multi-omics profiling of human vaccinees, we identify conserved early transcriptional and cellular signatures that predict subsequent adaptive responses across distinct adjuvant platforms, providing a framework for rational adjuvant selection.
