Decondensing cancer immunotherapy

Science

T cell activation predicates important immune responses. Programmed cell death protein 1 (PD-1) controls the overstimulation of T cells, and inhibiting PD-1 has been a successful strategy for immune-checkpoint cancer immunotherapy. Still, the precise mechanism by which PD-1 turns off T cells is not entirely clear. A better understanding could enable the design of more effective cancer drugs. Sharma et al. offer a new model for PD-1–mediated suppression of T cell function. Whereas other models focus on PD-1–mediated inhibitory effects on key signaling molecules in T cells, the authors propose that PD-1 dissolves biomolecular condensates (phase-separated droplet-like bodies) that are thought to amplify T cell signaling. The study offers a conceptually new framework for the negative regulation of T cell function that is potentially of therapeutic relevance.
