AMnESTI reverses age-associated lung immune decline to drive heterosubtypic protection by inactivated influenza vaccines

Science Immunology
Licensed inactivated influenza vaccines can protect children and adults against severe disease but are less effective in elderly populations. Gao et al. developed an adjuvant that combines manganese ions with an agonist of the cyclic GMP-AMP synthase–stimulator of interferon genes (cGAS-STING) pathway encapsulated in a liposome complex that can specifically target alveolar epithelial cells (AECs) in the lung. Intranasal delivery of this adjuvant promoted expansion of plasmacytoid dendritic cells and M1 macrophage polarization and revived immune responses in aged mice to levels comparable to those in young mice. A single dose of this adjuvant combined with inactivated influenza virus also conferred heterotypic immunity against a range of influenza strains.
