| 背景 | This is clone 2.4G2, a rat IgG2b monoclonal that recognises mouse CD32 (FcγRII) and CD16 (FcγRIII), produced in rat and offered as a low-endotoxin, in-vivo-ready preparation for functional studies. 2.4G2 is arguably the most widely used Fc-receptor blocking reagent in mouse immunology, so this format suits both benchtop and in-vivo applications: it blocks non-specific binding of labelled antibodies to Fcγ receptors in flow cytometry (the canonical "Fc block" step) and is also applied in vivo to interrogate the contribution of activating and inhibitory Fcγ receptors to antibody effector function, phagocytosis and immune-complex handling. Validated applications include Western blot, blocking, flow cytometry, IHC on frozen sections, immunoprecipitation and CODEX. Supplied bulk at milligram-to-gram scale with low endotoxin (<1 EU/mg research grade; <0.5 EU/mg ultra-low) to reduce confounding innate stimulation in animal work. CD32 (FcγRII) and CD16 (FcγRIII) are cell-surface receptors for the Fc region of IgG that translate antibody binding into cellular effector responses. In mouse, FcγRIII (CD16) and FcγRII (CD32) share a highly homologous extracellular region, which is why clone 2.4G2 recognises both. FcγRIII is an activating receptor that pairs with the ITAM-bearing FcRγ chain and drives antibody-dependent cellular cytotoxicity, phagocytosis and mediator release by NK cells, macrophages, neutrophils and mast cells. FcγRII (specifically FcγRIIb in mouse) is the single inhibitory receptor, carrying an ITIM that raises the threshold for activation and dampens B-cell and myeloid responses. The balance between these activating and inhibitory receptors sets the outcome of IgG-immune-complex engagement and is central to the effector activity of therapeutic and endogenous antibodies. Because 2.4G2 engages both, it is used to block Fcγ-receptor-mediated interactions broadly rather than to distinguish individual receptors. The Fc gamma II/III receptors are present on mouse natural killer cells, monocytes, macrophages, granulocytes, B-cells and most fetal thymocytes. |
