| 背景 | This product is a purified rat anti-mouse CD27 monoclonal antibody, clone RM27-3E5, of the rat IgG1 kappa isotype. Raised in rat against mouse CD27, it recognises the murine antigen and is validated for functional assays, flow cytometry and immunoprecipitation. The rat IgG1 backbone and defined clone make it well suited to in-vivo immunology and immuno-oncology studies in mouse models, where CD27 engagement is used to probe T-cell costimulation and antitumour immunity. The preparation is produced at low endotoxin levels (research grade below 1 EU/mg; ultra-low options below 0.5 EU/mg) to minimise confounding innate activation during animal dosing, and is supplied in bulk milligram-to-gram quantities appropriate for repeated-injection and multi-cohort experiments. All material is for research use only and is not intended for diagnostic or therapeutic applications. As with any functional-grade reagent, users should confirm activity, working concentration and lot suitability in their own system before scaling into full studies, and pair in-vivo work with appropriate isotype-matched control antibodies. CD27 (TNFRSF7; UniProt P41272) is a member of the tumour necrosis factor receptor superfamily and functions as a costimulatory receptor for T lymphocytes. It is constitutively expressed on most resting T cells, on subsets of NK cells and memory B cells, and its expression is upregulated following activation. Its single ligand is CD70 (TNFSF7), whose expression is tightly restricted to activated immune cells. Engagement of CD27 by CD70 recruits TRAF2 and TRAF5 adaptors, driving NF-kB and JNK signalling that promotes T-cell survival, clonal expansion, effector differentiation and the generation and maintenance of memory. CD27 costimulation supports both CD4 and CD8 responses and contributes to NK-cell and antibody responses. Because agonistic CD27 signalling can amplify antitumour T-cell immunity, the CD27-CD70 axis is an established target for costimulatory immunotherapy. CD27 is expressed on T cells, B cells, and NK cells | ||||
