| 背景 | This product is the rat anti-mouse CD24 monoclonal antibody, clone M1/69 (rat IgG2b, kappa), one of the most widely used and best-characterised CD24 clones in mouse immunology. It recognises the heat-stable antigen (HSA/CD24) and has been applied across flow cytometry, immunofluorescence, immunohistochemistry, immunoprecipitation and Western blot, making it a versatile tool for both phenotyping and functional work. This in-vivo formulation is manufactured to low endotoxin levels (research grade <1 EU/mg; ultra-low <0.5 EU/mg where specified) and supplied in bulk milligram-to-gram quantities suitable for antibody administration studies in mice, including tumour, immune-development and inflammation models. Because CD24 marks granulocytes, developing lymphocytes, many epithelial and neural precursors and a broad range of tumour cells, M1/69 is frequently used to track, sort or target these populations, and interest in CD24 as a myeloid "don't eat me" checkpoint has expanded its use in functional in-vivo experiments. All material is for research use only (RUO) and is intended as guidance for investigators designing their own studies. CD24, also called heat-stable antigen (HSA), is a small, heavily glycosylated glycosylphosphatidylinositol (GPI)-anchored cell-surface protein. Despite its short core peptide, extensive glycosylation makes it a versatile ligand and signalling partner. CD24 is expressed on granulocytes, developing B and T lymphocytes, many epithelial and neural progenitor cells, and is markedly upregulated on numerous carcinomas and cancer stem-like cells, where it is often used as a tumour marker. Functionally, CD24 modulates cell adhesion, proliferation and differentiation, and can serve as a ligand for P-selectin and for Siglec-10 (Siglec-G in mouse). The CD24-Siglec-10/G interaction transmits an inhibitory "don't eat me" signal that dampens macrophage phagocytosis and restrains inflammatory responses to damage-associated molecular patterns, positioning CD24 as an innate immune checkpoint. These roles make CD24 relevant to immune development, tissue homeostasis, autoimmunity and tumour immune evasion. CD24 is expressed on B cells, T cells, neutrophils, eosinophils, macrophages, neural cells, ganglion cells, keratinocytes, muscle cells, pancreas cells, lymphocytes, granulocytes, epithelial cells, thymocytes, monocytes, erythrocytes, dendritic cells and is overexpressed in many cancers. Expression varies during T and B cell differentiation. Peripheral T cells are mainly negative. | ||||
