| 背景 | This is a rat anti-mouse CD317 monoclonal antibody, clone 927, of the rat IgG2a kappa isotype, raised against mouse CD317 (BST-2/tetherin, also widely known as PDCA-1). The rat IgG2a backbone engages mouse activating Fc gamma receptors and complement, making the clone suitable not only for phenotyping but for antibody-mediated depletion and functional perturbation studies in vivo. Clone 927 is a well-recognized reagent for identifying and targeting plasmacytoid dendritic cells (pDCs), on which CD317/PDCA-1 is highly and characteristically expressed in the steady state. It is validated across flow cytometry, immunocytochemistry, and immunofluorescence microscopy, and is applied in depletion and functional assays. The product is manufactured to low-endotoxin specification and supplied in bulk milligram-to-gram quantities for animal work, avoiding the endotoxin-driven immune artifacts and volume limitations of standard research-grade lots. It is provided for research use only (RUO). These attributes make it appropriate for immunology studies of pDC biology, type I interferon responses, tumor immunology, and antiviral or autoimmune models where selective targeting of CD317-bearing cells is required. CD317, also called BST-2, tetherin, or PDCA-1, is a type II single-pass membrane protein with an unusual topology: an N-terminal cytoplasmic tail, a transmembrane domain, an extracellular coiled-coil ectodomain, and a C-terminal GPI anchor. This dual-membrane architecture underlies its best-characterized function as a restriction factor that physically tethers nascent enveloped virions to the surface of infected cells, inhibiting release of retroviruses and other enveloped viruses; it is strongly induced by type I interferon. Beyond antiviral restriction, CD317 acts as a signaling molecule and can modulate NF-kB activation. In the mouse, its most useful expression pattern for immunologists is its high, relatively selective presence on plasmacytoid dendritic cells in the steady state, which is why the PDCA-1 designation is used to identify and isolate pDCs. Expression is broadly inducible on many cell types under inflammatory or interferon-rich conditions. Murine CD317 is expressed by IFN-producing cells in naïve mouse spleen and by a wide variety of cell lines including T cells, mast cells, B cells, fibroblast cells, and pluripotent embryonal carcinoma cells. Additionally, mice challenged by influenza or other stimuli (CpG, LPS, murine CMV, poly(I:C), and imiquimod) express CD317 in DC and other myeloid cells as well as T cells, B cells, NKT cells, and some NK cells. CD317 is also expressed on CD138+ plasma cells in naïve mice and is upregulated by viral stimulation. | ||||
