RM134L is a rat IgG2b kappa monoclonal antibody raised against mouse CD252 (OX40 ligand, OX40L/TNFSF4). It recognises the mouse antigen and has been used for blocking, flow cytometry, frozen-section IHC, Western blotting and in vivo studies. As a rat IgG2b directed at a costimulatory ligand, it is well suited to functional and in vivo work: the format supports interruption of OX40-OX40L costimulation in intact animals and pairs naturally with flow-based readouts of activated T cells and OX40L-expressing antigen-presenting cells. This preparation is manufactured for in vivo use with low endotoxin (research grade <1 EU/mg; ultra-low-endotoxin option <0.5 EU/mg) to limit confounding innate immune activation, and is offered in bulk milligram-to-gram quantities for dosing across treatment groups and repeat-injection schedules. It is supplied for research use only (RUO). Because it targets the ligand rather than the OX40 receptor, RM134L is commonly selected when the experimental aim is to block the costimulatory interaction. It is expressed by activated antigen-presenting cells including dendritic cells, B cells and macrophages, and by endothelial cells, with expression induced upon activation. Its receptor is OX40 (CD134, TNFRSF4), which is upregulated on recently activated CD4+ and CD8+ T cells and on regulatory T cells. Engagement of OX40 by OX40L delivers a costimulatory signal that promotes T-cell survival, proliferation, effector differentiation and memory formation, and can modulate regulatory T-cell activity. The OX40-OX40L axis acts after initial priming to sustain and amplify T-cell responses and contributes to the pathogenesis of several inflammatory and autoimmune conditions, making the ligand an attractive target for interrupting excessive costimulation.
