| 背景 | Clone M/K-2.7 is a rat IgG1 kappa monoclonal antibody raised against mouse CD106 (VCAM-1). It is a widely used function-blocking anti-adhesion-molecule reagent, recognizing an epitope in the VLA-4-binding region and inhibiting VCAM-1-dependent leukocyte adhesion. The rat-anti-mouse format makes it well suited to in vivo studies in immunocompetent mouse models, where its neutralizing activity is used to interrupt endothelial and stromal adhesion cascades without depleting a cell population. This preparation is manufactured to low-endotoxin specification (research grade <1 EU/mg; ultra-low <0.5 EU/mg), which limits confounding TLR4-driven inflammation in adhesion, trafficking, and homing experiments where the readout is sensitive to background activation. Supplied in bulk milligram-to-gram quantities for repeat dosing and multi-cohort designs, and validated for immunofluorescence, in vivo administration, and neutralization applications. For research use only (RUO). All content here is provided as guidance for ichorbio to review before publication. CD106, better known as VCAM-1 (Vascular Cell Adhesion Molecule 1) and encoded by Vcam1 (UniProt P29533), is a type I transmembrane immunoglobulin-superfamily glycoprotein. It is induced on activated vascular endothelium by inflammatory cytokines such as TNF-alpha and IL-1, and is also expressed by bone marrow stromal cells, follicular dendritic cells, and some macrophages. Its principal counter-receptor is the integrin VLA-4 (alpha4-beta1) on lymphocytes, monocytes, and eosinophils, with additional binding to alpha4-beta7. Through VLA-4 engagement, VCAM-1 mediates firm adhesion and transendothelial migration of leukocytes during the multistep adhesion cascade, supporting recruitment to sites of inflammation. VCAM-1 also contributes to hematopoietic progenitor retention in bone marrow niches and to lymphocyte homing. It is a key player in atherosclerosis, autoimmune and allergic inflammation, and leukocyte trafficking, making it a frequent target for anti-adhesion intervention studies. |
