| 背景 | This is a bispecific in-vivo antibody combining the anti-mouse PD-L1 clone 10F.9G2 with the anti-mouse OX40 (CD134) clone OX-86, engineered on a mouse IgG2a, kappa framework carrying LALA-PG Fc mutations. The LALA-PG substitutions silence FcgammaR engagement and complement fixation, so activity is driven by simultaneous target binding rather than by Fc effector function. Pairing checkpoint blockade (PD-L1) with agonism of a costimulatory receptor (OX40) in a single molecule is a widely explored strategy to co-localise the two activities on the same cell or within the same microenvironment. Because both parental clones are among the most frequently used reagents for mouse PD-L1 blockade and OX40 agonism, the format is well suited to syngeneic tumour immunotherapy studies, T-cell activation experiments, and dual-mechanism combination modelling in immunocompetent mice. Supplied low-endotoxin, in vivo-ready, purified, and in scalable milligram-to-gram quantities for research use only. Note the product card target field reads "O+40," which appears to be a typo for OX40. PD-L1 (CD274, UniProt Q9EP73) is an inhibitory ligand expressed on tumour cells, myeloid cells, and other APCs. Engagement of its receptor PD-1 on T cells delivers a coinhibitory signal that dampens TCR-driven activation, promoting T-cell exhaustion and immune escape; blocking PD-L1 relieves this brake. OX40 (CD134, TNFRSF4, UniProt P47741) is a costimulatory member of the TNF receptor superfamily, transiently upregulated on activated CD4 and CD8 T cells and expressed on regulatory T cells. Its ligand OX40L (CD252) drives OX40 signalling that enhances effector T-cell proliferation, survival, and cytokine production and can destabilise or deplete intratumoural Tregs. Combining PD-L1 blockade with OX40 agonism is intended to simultaneously remove an inhibitory signal and deliver a positive costimulatory one, augmenting antitumour T-cell responses. |
