| 背景 | This bispecific in vivo antibody pairs two well-characterized mouse clones: TA99, which recognizes mouse TYRP1 (tyrosinase-related protein 1, P07147), and 145-2C11, which engages the mouse CD3 epsilon subunit of the T-cell receptor complex (P22646). Built on a mouse IgG2a kappa framework carrying LALA-PG mutations, its Fc is effector-silenced, so tumor-directed activity is driven by physical bridging of T cells to TYRP1-positive target cells rather than by Fc-mediated ADCC or complement. The mouse host origin and syngeneic reactivity make it well suited to fully immunocompetent mouse models, avoiding the anti-drug responses that limit non-mouse formats in chronic studies. Supplied at low endotoxin, in bulk milligram-to-gram quantities, and in an in-vivo-ready buffer, it is intended for functional and mechanistic work such as redirected T-cell cytotoxicity, melanoma-targeting T-cell engager studies, and tumor microenvironment interrogation. For research use only; not for diagnostic or therapeutic use. All content is provided as guidance for ichorbio review before publication. TYRP1 (tyrosinase-related protein 1) is a melanosomal membrane enzyme in the melanin biosynthesis pathway, expressed largely by melanocytes and melanoma cells. Its restricted, largely lineage-specific expression makes it an attractive tumor-associated target for redirecting immune effectors to melanoma while sparing most normal tissues. CD3 is the invariant signaling module of the T-cell receptor complex; the CD3 epsilon chain recognized by 145-2C11 is expressed on essentially all mature T cells. Engaging CD3 clusters the TCR complex and delivers an activating signal independent of peptide-MHC specificity. In a bispecific format, one arm anchors TYRP1 on the tumor cell while the other ligates CD3 on a passing T cell, forming an immunological synapse that triggers polyclonal T-cell activation and directed cytotoxicity against the TYRP1-positive target, regardless of the T cell's native antigen specificity. |
