| 背景 | FGL-1 (Fibrinogen-like protein 1, also known as HFREP1) is a secreted hepatokine of the fibrinogen family, produced primarily by hepatocytes and circulating in plasma. Beyond its roles in liver regeneration and metabolism, FGL-1 has emerged as a major immune checkpoint ligand: it binds LAG-3 on activated T cells and NK cells, and TACI on innate-like B cells, delivering inhibitory signals that dampen immune responses. FGL-1 is markedly upregulated in several tumor types (including lung, breast, and hepatocellular carcinoma), where it contributes to immune evasion within the tumor microenvironment, making it a target of active interest in cancer immunotherapy research. FGL-1 synthesized primarily by hepatocytes (liver); distributes systemically into plasma and adipose tissue. Low-level expression also reported in pancreas, skeletal muscle, and hematopoietic/reproductive tissues. It is also markedly upregulated in tumor tissue (lung, breast, liver cancers), where it acts as an inhibitory checkpoint ligand, binding LAG-3 on activated T cells (CD4+, CD8+, Tregs) and NK cells, and TACI on innate-like B cells. |
