Secukinumab is a fully human IgG1-kappa monoclonal antibody that binds and neutralizes interleukin-17A (IL-17A), a pro-inflammatory cytokine produced mainly by Th17 cells, gamma-delta T cells, and innate lymphoid cells. IL-17A signals through the IL-17RA/IL-17RC receptor complex on epithelial cells, keratinocytes, fibroblasts, and synoviocytes, driving expression of chemokines, antimicrobial peptides, and inflammatory mediators. Dysregulated IL-17A signaling contributes to psoriasis, psoriatic arthritis, and ankylosing spondylitis, the indications for which secukinumab is approved.
