背景

Natural killer (NK) cells are cytotoxic innate lymphocytes that mediate direct killing of infected and tumor cells and drive antibody-dependent cellular cytotoxicity (ADCC), with their functional state highly sensitive to ex vivo isolation method. The Ichorbio Untouched Human NK Cell Isolation Kit purifies human NK cells from PBMCs via column-based negative (untouched) selection using magnetic bead-conjugated antibodies, depleting non-target cells to yield untouched NK cells with purity up to 94.3%, free of surface-bound antibodies or beads and suitable for cytotoxicity, ADCC, and other functional assays requiring non-activated cells.

CD56 (NCAM) is the adhesion molecule characteristic of NK cells; CD16 (FcγRIII) is the low-affinity IgG receptor present on most peripheral NK cells; CD3 is used to actively exclude T lymphocytes, since some T cells (such as NKT cells) can also express CD56 or CD16.
