背景

T cells are the primary mediators of adaptive cellular immunity, encompassing CD4+ helper and CD8+ cytotoxic subsets that orchestrate antigen-specific responses, cytokine production, and cell-mediated cytotoxicity. As with other lymphocyte populations, their functional and activation state is highly sensitive to the isolation method used, making unmanipulated recovery essential for downstream functional assays. The Ichorbio Untouched Human Pan-T Cell Isolation Kit purifies human Pan-T cells from PBMCs via column-free negative (untouched) selection, using a biotinylated antibody cocktail and magnetic beads to label and sequester non-target cells, allowing untouched CD3+ T cells to be recovered directly from the supernatant. This yields untouched CD3+ T cells with purity up to 92.5%, free of surface-bound antibodies or beads and suitable for functional assays requiring non-activated, unmanipulated cells.

CD3 (as part of the TCR complex) is expressed on the surface of all mature T lymphocytes, from thymocyte subsets through peripheral T cells; absent on B cells, NK cells, and myeloid cells.
