Naïve CD8+ T cells (CD8+CD62L+CD44-) are antigen-inexperienced cytotoxic T lymphocytes that have not yet encountered cognate antigen, representing a critical starting population for studies of T cell priming, differentiation into effector or memory subsets, and antigen-specific cytotoxic responses. Their unactivated, quiescent state is highly sensitive to the isolation method used, making unmanipulated recovery essential for downstream functional and differentiation assays. The Ichorbio Untouched Mouse Naïve Cytotoxic T Cell Isolation Kit purifies naïve CD8+ T cells from mouse splenocytes or lymph node cells via column-free negative (untouched) selection, using a biotin-conjugated antibody cocktail and magnetic beads to label and sequester non-target and non-naïve cells, allowing untouched naïve CD8+ T cells to be recovered directly from the supernatant. This yields high-purity, untouched cells free of surface-bound antibodies or beads, with native phenotype and activation status preserved for sensitive downstream applications.

Cd8a is expressed on cytotoxic T lymphocytes and thymocyte subsets in mouse; naive CD8+ T cells are additionally CD62L-high/CD44-low, distinguishing them from memory/effector subsets.
