T cells are the primary mediators of adaptive cellular immunity, encompassing CD4+ helper and CD8+ cytotoxic subsets that orchestrate antigen-specific responses, cytokine production, and cell-mediated cytotoxicity. As with other lymphocyte populations, their functional and activation state is highly sensitive to the isolation method used, making unmanipulated recovery essential for downstream functional assays. The Ichorbio Untouched Mouse Pan-T Cell Isolation Kit purifies Pan-T cells from mouse splenocytes or lymph node cells via column-free negative (untouched) selection, using a biotin-conjugated antibody cocktail and magnetic beads to label and sequester non-target cells, allowing untouched CD3+ T cells to be recovered directly from the supernatant. This yields high-purity, untouched cells free of surface-bound antibodies or beads, with native phenotype and activation status preserved for sensitive downstream applications.

Cd3 (as part of the TCR complex) is expressed on all mature T lymphocytes in mouse; absent on B cells, NK cells, and myeloid cells.
