Removing this dominant population without disturbing the target cells' physiological state is essential for downstream functional and immunotherapy-related studies. The Ichorbio Indirect Human TCRαβ+ T Cell Depletion Kit depletes TCR alpha-beta+ cells from PBMCs via a column-based indirect magnetic labeling strategy, in which primary antibodies against TCR αβ are bound by anti-biotin magnetic nanobeads and retained in the separation column, allowing untouched non-TCR αβ cells (such as γδ T cells, NK cells, or B cells) to be recovered in the flow-through, achieving depletion efficiency up to 99.3%, yielding high-purity, untouched target cell populations free of antibody or bead residue and with zero activation, suitable for applications including γδ T cell and NK cell research, GvHD studies, and chimerism analysis. The alpha/beta T-cell receptor is expressed on the majority (~90-95%) of peripheral T lymphocytes, distinguishing alpha/beta T cells from the minor gamma/delta T-cell subset; absent on B cells, NK cells, and myeloid cells. |
