Prepared working solutions should be used on the day of preparation, and repeated freeze-thaw cycles of protein-containing reagents, including reconstituted standards/calibrators, should be avoided.

背景	Pacmilimab is a Probody therapeutic targeting PD-L1, engineered as a conditionally activated monoclonal antibody that remains masked in circulation and becomes activated within the tumor microenvironment through tumor-associated protease cleavage. This design aims to localize immune checkpoint blockade activity to tumors while reducing systemic immune-related toxicity. Once activated, pacmilimab blocks PD-L1 interaction with PD-1, restoring T-cell–mediated antitumor responses. The antibody has been evaluated in early clinical studies as a strategy to improve the therapeutic index of PD-L1 inhibition.
