This is recombinant human CD32a (FcgammaRIIa), clone/allotype designation 167H, produced in human embryonic kidney (HEK) 293 cells and supplied as an In Vivo Grade, ultra-low endotoxin (<0.5 EU/mg) preparation for research use only. As a HEK293-expressed human protein, it carries authentic mammalian folding and glycosylation, making it well suited to quantitative binding and interaction studies rather than animal dosing. The 167H designation refers to the histidine-167 allelic form of the FCGR2A extracellular region. It is characterised primarily in surface plasmon resonance (SPR), biolayer interferometry (BLI) and ELISA formats, where it is used to measure the affinity of IgG Fc domains, therapeutic antibodies and immune complexes for this low-affinity activating Fc receptor. Ultra-low endotoxin and bulk mg-to-gram availability support reagent-intensive workflows such as kinetic screening, epitope/Fc-engineering campaigns and effector-function profiling. Note: this is a soluble recombinant protein reagent, not a functional antibody; it has no depleting, blocking or agonist mechanism of its own. CD32a (FcgammaRIIa), encoded by FCGR2A, is a low-affinity activating receptor for the Fc region of immunoglobulin G. It is broadly expressed on myeloid and phagocytic cells including monocytes, macrophages, neutrophils, dendritic cells and platelets. Unlike most Fcgamma receptors, CD32a signals through an intrinsic cytoplasmic ITAM (immunoreceptor tyrosine-based activation motif) rather than a separate gamma chain, triggering phagocytosis, degranulation, respiratory burst and cytokine release upon crosslinking by IgG-opsonised targets or immune complexes. It engages IgG1 and IgG3 most strongly. A well-known functional polymorphism at position 131 (histidine versus arginine) alters binding to human IgG2 and influences responses to therapeutic antibodies. CD32a is central to antibody-dependent cellular phagocytosis and immune-complex clearance, making it a key determinant of monoclonal antibody effector function and a frequent target in Fc engineering.
