| 稳定性和存储 | Lyophilized proteins are stable at ambient temperature for at least 2 weeks. If the protein is not to be used immediately then the protein should be stored in lyophilized form at -20 °C for up 12 months. Once the protein has been reconstituted we recommend storage at 4 °C for up to one week. For longer term storage of protein in solution we recommend aliquoting into smaller vials to avoid repeated freeze-thaw cycles and storage at -20 or -80 °C for up to 3 months. To avoid surface adsorption loss and inactivation we strongly recommend that the protein should not be aliquoted in less than 10 μg per vial. |

| 背景 | This is a recombinant human neonatal Fc receptor (FcRn) heterodimer protein produced in HEK293 cells, comprising the FcRn alpha chain (FCGRT, UniProt P55899) non-covalently associated with beta-2-microglobulin (B2M, UniProt P61769). Expression in a mammalian system supports proper folding and physiologically relevant post-translational modifications. It is supplied as an in-vivo-grade, ultra-low-endotoxin (<0.5 EU/mg) preparation suitable for demanding functional and biophysical studies, and is available in bulk (mg to gram) quantities. The protein is validated for surface plasmon resonance (SPR), bio-layer interferometry (BLI), and ELISA, making it well suited to characterizing pH-dependent IgG and albumin binding, screening Fc-engineered antibodies and half-life-extended biologics, and ranking candidate molecules by their FcRn interaction. As a heterodimer rather than the alpha chain alone, it more faithfully reproduces the native binding interface. All material is for research use only (RUO). The neonatal Fc receptor (FcRn) is an MHC class I-like molecule composed of an FCGRT alpha chain paired with beta-2-microglobulin. It is the master regulator of IgG and serum albumin homeostasis. FcRn binds the Fc region of IgG and albumin in a strictly pH-dependent manner: tight binding occurs in the acidic environment of the endosome (around pH 6.0), while binding is released at the near-neutral pH of the extracellular space and blood (pH 7.4). This behavior underlies FcRn-mediated recycling, whereby pinocytosed IgG and albumin are diverted away from lysosomal degradation and returned to the circulation, dramatically extending their serum half-lives. FcRn also mediates transcytosis of IgG across epithelial and endothelial barriers, including maternal-fetal transfer of immunity. Because of this central role, FcRn is a key determinant of the pharmacokinetics of antibody therapeutics and the target of Fc-engineering strategies and FcRn-antagonist drugs. |
