To obtain a final concentration of 1 mg/ml reconstitute 0.25 mg vials with 0.25 ml water and 1.0 mg vials with 1.0 ml water. Please refer to the specific buffer information in the hardcopy of datasheet or the lot-specific COA. |
| 运输 | Ambient |
| 稳定性和存储 | Lyophilized proteins are stable at ambient temperature for at least 2 weeks. If the protein is not to be used immediately then the protein should be stored in lyophilized form at -20 °C for up 12 months. Once the protein has been reconstituted we recommend storage at 4 °C for up to one week. For longer term storage of protein in solution we recommend aliquoting into smaller vials to avoid repeated freeze-thaw cycles and storage at -20 or -80 °C for up to 3 months. To avoid surface adsorption loss and inactivation we strongly recommend that the protein should not be aliquoted in less than 10 μg per vial. |

| 背景 | This product is a recombinant mouse CD16a (Fc gamma receptor III, FcgRIII) protein, supplied as the soluble extracellular domain expressed in human embryonic kidney (HEK) 293 cells. Mammalian expression supports native-like folding, disulfide bonding, and glycosylation, which are important for retaining functional IgG-Fc binding activity in solution. It is manufactured to an ultra-low endotoxin specification (<0.5 EU/mg), making it well suited to sensitive functional and cell-based assays where endotoxin can confound readouts. The protein is provided in bulk milligram-to-gram quantities for research use only (RUO). Note that this is a soluble recombinant protein reagent, not a monoclonal antibody: it carries no clone designation, isotype, or host-species immunoglobulin framework. Its stated applications are in-vitro binding and biophysical characterization methods, specifically surface plasmon resonance (SPR), bio-layer interferometry (BLI), and ELISA. Typical uses include measuring Fc-receptor engagement of therapeutic antibody candidates, ranking Fc-engineering variants, and studying affinity/kinetics of the FcgR-IgG interaction (UniProt P08508). CD16a (Fc gamma receptor III, FcgRIII; UniProt P08508) is a low-to-moderate affinity receptor for the Fc region of IgG and a key activating Fc receptor in mice. It is expressed on natural killer (NK) cells, macrophages, monocytes, and other myeloid populations, where it recognizes IgG immune complexes and antibody-opsonized targets. Engagement clusters the receptor and signals through associated immunoreceptor tyrosine-based activation motif (ITAM)-bearing adaptor chains (the FcR gamma chain), triggering effector programs. Functionally, CD16a is central to antibody-dependent cellular cytotoxicity (ADCC) and antibody-dependent cellular phagocytosis (ADCP), linking humoral immunity to cellular effector responses. Because it drives the effector arm of many therapeutic antibodies, the affinity and kinetics of IgG-Fc binding to CD16a are important determinants of in-vivo antibody activity, and recombinant CD16a ectodomain is widely used to characterize these interactions. |
