| 稳定性和存储 | Lyophilized proteins are stable at ambient temperature for at least 2 weeks. If the protein is not to be used immediately then the protein should be stored in lyophilized form at -20 °C for up 12 months. Once the protein has been reconstituted we recommend storage at 4 °C for up to one week. For longer term storage of protein in solution we recommend aliquoting into smaller vials to avoid repeated freeze-thaw cycles and storage at -20 or -80 °C for up to 3 months. To avoid surface adsorption loss and inactivation we strongly recommend that the protein should not be aliquoted in less than 10 μg per vial. |

| 背景 | This is a recombinant marmoset (Callithrix jacchus) CD16a (FcgammaRIIIa) protein produced in human embryonic kidney (HEK) 293 cells, which supports mammalian folding and glycosylation appropriate for a native Fc receptor. It is supplied as an In Vivo Grade, ultra-low endotoxin preparation (<0.5 EU/mg), making it suitable for sensitive functional, cell-based and receptor-interaction studies where endotoxin contamination would confound results. The protein is validated for label-free binding and immunoassay applications including surface plasmon resonance (SPR), bio-layer interferometry (BLI) and ELISA, and is well suited to measuring IgG-Fc/FcgammaRIIIa binding kinetics and affinity. As a non-human primate ortholog, it is a useful reagent for cross-species characterization of therapeutic antibody Fc engagement, Fc-engineering and glycoengineering campaigns, and comparative effector-function studies that bridge rodent and human systems. Material is offered as a research-use-only (RUO) reagent available in bulk quantities. UniProt: A0A8I3WA45. Note this is a soluble recombinant protein reagent, not an antibody.
CD16a (FcgammaRIIIa) is a low-affinity activating receptor for the Fc region of IgG, and the marmoset ortholog belongs to the same FcgammaRIII/CD16 family conserved across primates. It is expressed principally on natural killer cells, and also on monocytes, macrophages and subsets of other myeloid cells. On NK cells, CD16a associates with ITAM-bearing signaling adaptors (such as the FcRgamma chain and/or CD3zeta) and, upon engaging antibody-opsonized target cells, triggers antibody-dependent cellular cytotoxicity (ADCC) and cytokine release. Because CD16a directly couples the specificity of an IgG antibody to NK-mediated killing, it is a central effector-arm receptor for therapeutic antibodies and a determinant of their in-vivo potency. The marmoset (Callithrix jacchus) form is of particular interest for translational and cross-species Fc-engineering work, where non-human primate receptors help predict human effector-function behavior. |
