| 背景 | This antibody is a recombinant version of clone RMP1-14, a rat-derived anti-mouse PD-1 (CD279) specificity that has become one of the most widely used tools for immune checkpoint blockade in preclinical mouse models. It recognises mouse PD-1 (UniProt Q02242) and blocks the interaction between PD-1 and its ligands PD-L1 and PD-L2, thereby relieving inhibitory signalling in T cells. Reformatting the clone as a recombinant antibody gives sequence-defined, lot-to-lot consistency compared with hybridoma-derived material, an important consideration for reproducible functional and in-vivo studies. Supplied on a mouse IgG2a scaffold, it is validated for Western blot, blocking, flow cytometry and functional assays. It is manufactured to be in-vivo-ready with low endotoxin and is available in bulk milligram-to-gram quantities, making it suitable for immuno-oncology models, tumour immunology, T-cell exhaustion studies and combination-therapy experiments. The product is for research use only (RUO) and is not intended for diagnostic or therapeutic use. PD-1 (Programmed cell death protein 1, CD279) is an inhibitory receptor of the CD28/CTLA-4 immunoglobulin superfamily, expressed on activated T cells, B cells, natural killer cells and other immune subsets. Its cytoplasmic tail contains ITIM and ITSM motifs that recruit the phosphatase SHP-2 upon ligand engagement, dampening TCR and CD28 signalling. Its two ligands, PD-L1 and PD-L2, are found on antigen-presenting cells, many tumour cells and inflamed tissue. Under physiological conditions this axis maintains peripheral tolerance and limits collateral tissue damage during immune responses. In chronic infection and cancer, sustained PD-1 expression marks exhausted, dysfunctional T cells, and tumours exploit PD-L1 upregulation to evade immune attack. Blocking the PD-1/PD-L axis can restore effector T-cell function, which is the rationale underlying checkpoint-inhibitor immunotherapy.
