Clone 10F.9G2 is a widely used mouse anti-mouse PD-L1 (CD274/B7-H1) monoclonal antibody, offered here as a recombinant mouse IgG1 that reproduces the original clone's binding specificity with defined, lot-to-lot consistent production. Its reactivity against mouse PD-L1 and its ability to block the PD-1/PD-L1 interaction make it well suited to functional immunology and in-vivo checkpoint-blockade studies in syngeneic tumour and chronic infection models, as well as flow cytometry, IHC on frozen sections, immunofluorescence, and in-vitro functional assays. Supplied at low endotoxin (research-grade <1 EU/mg, ultra-low <0.5 EU/mg) and available in bulk milligram-to-gram quantities, it is intended to support repeat-dosing regimens and larger cohorts without switching lots. PD-L1 (programmed death-ligand 1; also called CD274 or B7-H1) is a type I transmembrane glycoprotein of the B7 family and a principal ligand for the inhibitory receptor PD-1. Engagement of PD-1 by PD-L1 recruits SHP-2 phosphatase to the PD-1 cytoplasmic tail, dampening TCR and CD28 signalling and thereby attenuating T-cell proliferation, cytokine production, and cytotoxic activity. PD-L1 is expressed on many immune cells and non-haematopoietic tissues and is strongly upregulated by interferon-gamma, allowing tumours and chronically infected tissues to co-opt this pathway to evade immune attack. PD-L1 also binds CD80 (B7-1) in cis and trans, adding further regulatory layers. Because the PD-1/PD-L1 axis is a central mechanism of peripheral tolerance and adaptive immune resistance, blocking it is a cornerstone strategy for restoring effector T-cell function in cancer and infection models.
