To detect Mouse PD-1 this antibody can be used at a concentration of 30 µg/ml.
Blocking: The 29F.1A12 antibody has been shown to block the binding of PD-1 to its ligands in vivo. For immune checkpoint antibodies 200 ug per mouse per injection is the standard dosage in the literature. NOTE: The response is model dependent. We have seen some tumor lines in mice that grow progressively. If we treat the animal with anti-PD-1 (days 3,6,9 @ 200 ug per animal per injection) then the tumor will regress, and disappear.
Each investigator should determine their own optimal working dilution for specific applications. |

| 背景 | Clone 29F.1A12 is a mouse IgG2a monoclonal antibody directed against mouse PD-1 (PDCD1), supplied as a recombinantly produced reagent for reproducible lot-to-lot performance. Because it recognises and blocks the mouse ortholog, it is well suited to syngeneic tumour models and other immuno-oncology and chronic-infection studies in mice, where PD-1/PD-L1 checkpoint interference is a central experimental question. It is validated for flow cytometry, western blot, blocking assays, and IHC on frozen sections, supporting both phenotyping and functional work. ichor.bio offers this clone in a low-endotoxin (<1 EU/mg research grade; <0.5 EU/mg ultra-low) format at bulk milligram-to-gram scale, which is the practical requirement for repeated intraperitoneal dosing across treatment cohorts. The recombinant, in-vivo-ready presentation makes it appropriate for functional PD-1 blockade experiments as well as ex-vivo characterisation. All material is for research use only. The clone is a commonly cited tool for interrogating PD-1-dependent T-cell regulation in preclinical settings.
PD-1 (programmed cell death protein 1; PDCD1; UniProt Q02242) is an inhibitory immune checkpoint receptor of the CD28/CTLA-4 immunoglobulin superfamily, expressed on activated T cells, B cells, NK cells, and myeloid subsets. It is induced upon antigen engagement and is sustained at high levels on chronically stimulated and exhausted T cells. Engagement by its ligands PD-L1 (CD274) and PD-L2 (PDCD1LG2) recruits the phosphatases SHP-2 (and SHP-1) to PD-1's cytoplasmic ITIM and ITSM motifs, dampening TCR and CD28 signalling and attenuating proliferation, cytokine output, and cytotoxicity. This axis maintains peripheral tolerance and limits immunopathology, but tumours and persistent pathogens co-opt it to suppress effector responses.
