| 背景 | This is an engineered bispecific monoclonal antibody that simultaneously targets mouse PD-L1 (via the 10F.9G2 binding arm) and mouse CD47 (via the A4 binding arm), formatted on a mouse IgG2a, kappa backbone carrying LALA-PG Fc mutations to silence FcgammaR engagement and complement fixation. Because both parent clones are among the most widely used, well-characterised antibodies for blockade of their respective targets in mouse models, the bispecific pairing is well suited to in-vivo and functional studies of combined adaptive (T-cell) and innate (macrophage) checkpoint modulation within a single reagent. The murine framework and effector-silenced Fc minimise host anti-antibody responses and confounding effector-mediated depletion during repeat dosing in immunocompetent mice, making it appropriate for syngeneic tumour and mechanistic immunology work. Supplied as a low-endotoxin (research grade typically <1 EU/mg; ultra-low <0.5 EU/mg), in-vivo-ready preparation available in bulk milligram-to-gram quantities. For research use only (RUO); it is not for diagnostic or therapeutic use. Endotoxin grade, buffer, and exact isotype/format should be confirmed against the specific lot documentation before study design. PD-L1 (CD274, B7-H1; UniProt Q9EP73) is a B7-family transmembrane ligand expressed on tumour cells, antigen-presenting cells, and stromal cells. Engagement of its receptor PD-1 on T cells delivers an inhibitory signal that dampens TCR-driven proliferation and cytokine production, a pathway tumours co-opt to escape adaptive immunity. CD47 (UniProt Q61735) is a broadly expressed cell-surface protein that binds SIRPalpha on macrophages and dendritic cells, transmitting a "don't eat me" signal: SIRPalpha ITIM phosphorylation recruits SHP-1, suppressing cytoskeletal rearrangement and phagocytosis. Many tumours overexpress CD47 to evade macrophage-mediated clearance. Co-targeting both axes links relief of adaptive T-cell suppression (PD-L1/PD-1) with restoration of innate phagocytic clearance (CD47/SIRPalpha), a rationale behind bispecific PD-L1 x CD47 strategies designed to concentrate CD47 blockade at PD-L1-positive tumour cells while limiting on-target binding to normal tissues. |
