| 背景 | This is a fully mouse bispecific antibody engineered to bridge two clinically validated targets in a single molecule: the CD3 epsilon chain of the T-cell receptor complex (clone 145-2C11) and the B-cell antigen CD20 (clone MB20-11). Presented on an IgG2a, kappa backbone carrying LALAPG mutations, the Fc region is effector-silenced, so activity is driven by antigen bridging rather than by Fc gamma receptor or complement engagement. This design makes it well suited to in-vivo and functional studies of CD3-based T-cell redirection against CD20-positive B cells in immunocompetent mouse models, where a fully mouse-reactive, Fc-silent surrogate is required to model human T-cell engager therapeutics without confounding effector function. Supplied as a low-endotoxin, bulk, research-grade reagent (RUO), it is intended for repeat-dosing in-vivo experiments, ex-vivo T-cell activation and B-cell cytotoxicity assays, and mechanistic work on immune synapse formation, cytokine dynamics, and B-cell depletion kinetics. Researchers typically search for it by the component clone names 145-2C11 and MB20-11. CD3 is the invariant signaling module of the T-cell receptor (TCR) complex; its epsilon, gamma, delta and zeta chains transduce antigen recognition into intracellular signaling via ITAM phosphorylation, driving T-cell activation, proliferation and effector function. Antibodies to CD3 epsilon (such as clone 145-2C11) can crosslink and activate T cells independently of peptide-MHC. CD20 is a B-lymphocyte surface phosphoprotein expressed from the pre-B through mature B-cell stages but absent on plasma cells and stem cells; it functions as a store-operated calcium channel component and organizes membrane microdomains during B-cell activation. Because CD20 is B-lineage restricted and not shed or internalized appreciably, it is a durable target for B-cell directed therapy. A CD3 x CD20 bispecific physically bridges a cytotoxic T cell to a CD20-positive B cell, forming an artificial immune synapse that triggers polyclonal, MHC-independent T-cell activation and redirected killing of the B cell, independent of the T cell's native antigen specificity. |
