Anti-Human PD-L1 clone IB6 is a humanized IgG1 kappa monoclonal antibody recognizing human PD-L1 (CD274, B7-H1; UniProt Q9NZQ7). As a humanized IgG1 with intact effector-capable framework, it is suited to functional immuno-oncology studies where blockade of the PD-1/PD-L1 axis is the primary readout, and it can also engage Fc-dependent effector functions against PD-L1-positive target cells. PD-L1 (CD274, B7-H1; UniProt Q9NZQ7) is a type I transmembrane protein of the B7 immunoglobulin superfamily and the principal ligand for the inhibitory receptor PD-1 on T cells. Engagement of PD-1 by PD-L1 delivers a co-inhibitory signal that dampens T-cell receptor signalling, reduces effector cytokine production and proliferation, and promotes T-cell exhaustion, contributing to peripheral tolerance. PD-L1 is expressed on antigen-presenting cells and many non-haematopoietic tissues, and is frequently upregulated on tumour cells and myeloid cells, often driven by interferon-gamma. This adaptive expression allows tumours to co-opt the pathway to evade immune attack. PD-L1 also binds CD80 (B7-1), adding regulatory complexity. Because it is the tumour-side ligand of the axis, PD-L1 is a central target for checkpoint blockade aimed at restoring anti-tumour T-cell activity.
