| 背景 | Anti-Human BCMA (clone IB10) is a recombinant fully human IgG1 monoclonal antibody targeting human B-cell maturation antigen (BCMA/TNFRSF17, UniProt Q02223). As a human IgG1 reactive against a human antigen, it is well suited to in-vitro functional and mechanistic studies, target-validation work, and as a building block or benchmark reagent in therapeutic antibody development programmes rather than to conventional mouse in-vivo tumour dosing. The recombinant human IgG1 format provides lot-to-lot consistency, defined sequence, and an intact human Fc capable of engaging human Fc gamma receptors and complement, making it appropriate for ADCC/ADCP/CDC assays, binding and blocking studies, flow cytometry, and screening against BCMA-expressing plasma cells and multiple myeloma lines. Supplied at low endotoxin and available in bulk milligram-to-gram quantities, it supports assay-intensive and reagent-heavy workflows where endotoxin-driven artefacts must be minimised. BCMA (B-cell maturation antigen), also known as TNFRSF17 or CD269, is a member of the TNF receptor superfamily encoded by TNFRSF17. Its expression is largely restricted to the B-cell lineage, becoming prominent as B cells differentiate into plasmablasts and long-lived plasma cells, while being low or absent on naive and memory B cells. BCMA binds two TNF-family ligands, APRIL (high affinity) and BAFF (lower affinity), triggering NF-kB, PI3K/AKT, and MAPK signalling that promotes plasma-cell survival and antibody-secreting function. Because expression is high and consistent on malignant plasma cells, BCMA is a leading target in multiple myeloma, exploited by CAR-T cells, bispecific T-cell engagers, and antibody-drug conjugates. Soluble BCMA, shed by gamma-secretase, circulates and can serve as a disease biomarker and a potential ligand sink affecting targeting. |
