Mesothelin (MSLN) is a GPI-anchored cell-surface glycoprotein expressed at low levels on normal mesothelial cells lining the pleura, peritoneum, and pericardium, but strongly overexpressed in many epithelial malignancies including mesothelioma, pancreatic ductal adenocarcinoma, ovarian carcinoma, and certain lung and cholangiocarcinomas. The protein is synthesized as a precursor that is cleaved to release megakaryocyte-potentiating factor and the membrane-bound mature mesothelin. Mesothelin binds the ovarian cancer antigen MUC16 (CA-125), an interaction implicated in cell adhesion and peritoneal metastatic spread, and signalling associated with mesothelin has been linked to proliferation and resistance to apoptosis in some tumour contexts. Its restricted normal-tissue distribution combined with high tumour expression has made mesothelin a prominent target for antibody-drug conjugates, immunotoxins, bispecific antibodies, and CAR-T cell approaches, and a frequent antigen in translational immuno-oncology research. |
