Anti-Human CD25 (IB7) is a recombinant human IgG1 kappa monoclonal antibody directed against the human interleukin-2 receptor alpha chain (CD25, IL2RA; UniProt P01589). As a fully human-framework IgG1, clone IB7 is well suited to research applications that require engagement of the human target, including in-vitro functional assays, receptor-blocking and binding studies, flow cytometry, and mechanistic work on IL-2 signalling and regulatory T-cell biology. Because the reactivity is human, mouse in-vivo depletion or blockade regimens do not translate directly; the antibody is most appropriate for human cell systems, humanised models, or engineered surrogate settings. Supplied at research-grade low endotoxin (<1 EU/mg; ultra-low <0.5 EU/mg options) and available in bulk milligram-to-gram quantities, it is manufactured for reproducible, high-input experimental use. CD25 is the alpha chain of the interleukin-2 receptor (IL2RA). It is a non-signalling subunit that, together with the IL-2 receptor beta (CD122) and common gamma (CD132) chains, forms the high-affinity trimeric IL-2 receptor. CD25 itself does not transduce signal but greatly increases the receptor's affinity for IL-2, enabling responsiveness at low cytokine concentrations. CD25 is constitutively expressed at high levels on regulatory T cells (Tregs), where IL-2 signalling supports FOXP3 expression, survival, and suppressive function, and is transiently upregulated on activated effector T and B cells. Through the high-affinity receptor, IL-2 drives JAK1/JAK3 activation and STAT5 phosphorylation, influencing T-cell proliferation, differentiation, and tolerance. Because of its restricted, activation- and Treg-associated expression, CD25 is a widely studied marker and functional target in immune activation, autoimmunity, transplantation tolerance, and tumour immunology.
