| 背景 | IchorDetect Goat anti-Mouse IgG (H&L) F(ab')2 Secondary Antibody, PE (cat ICH3037), is a phycoerythrin-conjugated secondary detection reagent, not a primary antibody, biosimilar, or in-vivo therapeutic. It is raised in goat against whole-molecule mouse IgG, so it recognises both the heavy and light chains (H&L) and therefore binds all mouse IgG subclasses as well as, via shared light chains, other mouse immunoglobulin classes. The reagent is supplied as an F(ab')2 fragment, meaning the Fc region has been removed by pepsin digestion; this reduces non-specific binding to Fc receptors on live cells, macrophages, and other FcR-bearing populations, which is useful in flow cytometry and immunofluorescence on unfixed samples. It is cross-adsorbed against immunoglobulins or serum proteins of other common species to minimise cross-reactivity and background when multiplexing or working in mixed-species systems. PE is a bright, single-molecule fluorophore excited by the 488 or 561 nm laser lines with emission near 578 nm, making it well suited to flow cytometry and fluorescence microscopy. This is a research-use-only detection tool for indirect labelling of mouse primary antibodies.
The target here is mouse immunoglobulin G recognised across both heavy and light chains (H&L). IgG is the most abundant serum immunoglobulin and the workhorse isotype for most mouse monoclonal and polyclonal primary antibodies used in research. Because this secondary is anti-H&L, it detects epitopes on the gamma heavy chain and on the kappa/lambda light chains, so it will bind essentially any mouse IgG primary regardless of subclass, and can also detect other mouse Ig classes that share light chains. The F(ab')2 format removes the Fc portion of the secondary itself, lowering the chance of it engaging Fc gamma receptors on the sample. Cross-adsorption removes antibody sub-populations that would otherwise react with immunoglobulins from unrelated host species, which is important when a mouse primary is used alongside primaries raised in rabbit, rat, or human, or when staining tissue from those species. |
