背景

Classical monocytes (CD14+CD16-) are the predominant monocyte subset in peripheral blood, serving as precursors for macrophages and dendritic cells and playing key roles in innate immunity, phagocytosis, and inflammatory responses. Their differentiation potential and functional state are highly sensitive to the isolation method used, making unmanipulated recovery essential for downstream applications such as macrophage differentiation. The Ichorbio Untouched Human Classical Monocyte Isolation Kit purifies human CD14+CD16- classical monocytes from PBMCs via column-free negative (untouched) selection, using a biotin-conjugated antibody cocktail and magnetic beads to label and sequester non-target cells, allowing untouched classical monocytes to be recovered directly from the supernatant. This yields high-purity, untouched cells free of surface-bound antibodies or beads, with high viability and native phenotype preserved for sensitive downstream applications.

CD14 is expressed on classical (CD14++CD16-) monocytes and macrophages, and at lower levels on granulocytes and dendritic cells; a canonical marker of the monocyte/macrophage lineage.
