The CD4+ Helper T-Cell Depletion and Validation Kit (ICH8264) is a paired-reagent set designed for controlled depletion of CD4+ helper T cells in mouse models and independent confirmation of that depletion by flow cytometry. Both listed components are Rat IgG2b, Kappa antibodies reactive against mouse CD4, consistent with a kit format pairing a depleting anti-CD4 clone with a non-competing detection/validation clone so that residual CD4+ cells can be tracked without epitope masking by the depleting antibody. Reagents are supplied research grade (RUO), typically low endotoxin and formulated for in-vivo administration and bulk use. This kit-style product is intended to standardise depletion experiments: the depleting antibody removes the CD4 compartment while the validation antibody, binding a distinct epitope, enables accurate quantification of depletion efficiency in blood, spleen, or lymph node. CD4 is a single-chain transmembrane glycoprotein of the immunoglobulin superfamily expressed on helper T cells, and at lower levels on some monocytes, macrophages, and dendritic cells. It acts as a co-receptor for the T-cell receptor, binding the non-polymorphic beta-2 region of MHC class II molecules on antigen-presenting cells and recruiting the tyrosine kinase Lck to stabilise the immunological synapse and amplify TCR signalling. CD4+ helper T cells orchestrate adaptive immunity by providing help to B cells, licensing dendritic cells, and secreting polarising cytokines (Th1, Th2, Th17, Tfh, and regulatory subsets). Because of this central coordinating role, antibody-mediated depletion of CD4+ cells is a widely used approach to dissect helper T-cell contributions to infection, autoimmunity, transplantation, and antitumour immunity in mouse models.
