The Neutrophil Depletion & Validation Kit (ICH8267) is a paired-reagent set for depleting neutrophils in mice and confirming the extent of that depletion by flow cytometry. The stated isotypes (Rat IgG2a and Rat IgG2b kappa) are consistent with a depleting anti-Ly6G component (the widely used clone 1A8 is a rat IgG2a that selectively targets Ly6G on neutrophils) supplied together with a validation/detection antibody used to enumerate the remaining neutrophil compartment without simply re-detecting the depleting clone. Neutrophils are the most abundant circulating granulocytes and the first responders of innate immunity, rapidly recruited to sites of infection and injury where they phagocytose pathogens, degranulate, generate reactive oxygen species and release neutrophil extracellular traps. In mice the canonical surface handle for neutrophils is Ly6G, a GPI-anchored member of the Ly6 family expressed on mature granulocytes; the closely related Ly6C is shared with monocytes, which is why the older Gr-1 (RB6-8C5) reagent, recognising both Ly6G and Ly6C, depletes monocytes as well and has been largely superseded by Ly6G-selective clones such as 1A8 for specific neutrophil targeting. Neutrophils also express myeloid markers including CD11b and CD11b/Ly6G combinations are routinely used to gate them by flow cytometry.
