生物活性	AGS-1C4D4 displays potent ADCC activity against PSCA-expressing HPAC and Panc0203 pancreatic cell lines, with activity greatly reduced by deglycosylation, and is more potent than the hybridoma-derived AGS-PSCA antibody in this assay [1]. AGS-1C4D4 exhibits CDC activity against PSCA-expressing recombinant B300.19 cells, with activity greatly reduced by deglycosylation, and is equally effective as the hybridoma-derived AGS-PSCA antibody in this assay [1]. AGS-1C4D4 binds to PSCA with a K d of 0.2 nM, displaying similar binding affinity to the hybridoma-derived AGS-PSCA antibody [1]. AGS-1C4D4 significantly inhibits established pancreatic tumor growth and metastasis in orthotopic HPAC xenograft models when combined with Gemcitabine (), and also significantly inhibits pancreatic tumor formation, local invasion, and distant metastasis in the same orthotopic xenograft model [1].

Flow cytometry

Flow-cytometry using anti-human PSCA antibody. HPAC cells were stained with an irrelevant antibody (Blue Histogram) or an anti-human PSCA monoclonal antibody (Catalog HT361016, Yellow Histogram) at a concentration of 5 µg/ml for 30 mins at RT. After washing, bound antibody was detected using a a Goat Anti-Human IgG H&L Polyclonal Antibody, FITC (abinScience: HF690414) and cells analysed on a NovoCyte Flow Cytometer.
