| 背景 | The CAR Modulation Antibody Panel (ICH9003) is a curated set of six research biosimilars assembled to support chimeric antigen receptor (CAR) T-cell and adoptive cell therapy workflows. Each member is an unconjugated, non-therapeutic analog built around the target of a well-characterised originator antibody and supplied for research use only: an anti-CD3 (muromonab/OKT3-type) and an anti-CD28 (theralizumab-type) for T-cell activation and expansion; an anti-4-1BB/CD137 (urelumab-type) as a costimulatory agonist; anti-PD-1 (pembrolizumab-type) and anti-PD-L1 (atezolizumab-type) for checkpoint-axis studies; and an anti-TGF-beta (fresolimumab-type) to probe suppressive signalling in the tumour microenvironment. Together the panel spans the core signals that govern engineered T-cell priming, costimulation, exhaustion and immunosuppression. Reagents are manufactured to in-vivo research grade with low endotoxin, are RUO, and are available in bulk mg-to-g quantities. They are intended as functional tools and controls, not as clinical drugs. Because the panel bundles multiple distinct molecules, each component should be validated independently for the user's specific assay and target. The panel targets the signalling nodes that shape engineered T-cell behaviour. CD3 (part of the TCR complex) delivers signal 1 upon crosslinking, driving activation. CD28 provides signal 2 costimulation; its superagonist targeting (theralizumab-type) can drive strong polyclonal expansion. 4-1BB/CD137, a TNF-receptor-superfamily costimulator, sustains T-cell survival and memory and is a common CAR intracellular domain. PD-1 and its ligand PD-L1 form the dominant inhibitory checkpoint axis that restrains activated and exhausted T cells; blockade of either relieves suppression. TGF-beta is a pleiotropic cytokine that enforces immunosuppression, Treg induction and stromal remodelling in the tumour microenvironment. Studying these targets together lets investigators dissect activation, costimulation, exhaustion and microenvironmental suppression that determine CAR-T persistence and potency. | ||||
