什么是辅助T细胞?

辅助T细胞(Helper T cells, Th cells)是免疫系统中最重要的淋巴细胞亚群之一,主要由 CD4+ T细胞构成。它们在免疫应答中起到指挥官的作用,通过分泌细胞因子、帮助B细胞分泌抗体、促进巨噬细胞杀伤病原体、调节其他免疫细胞功能等方式,协调机体的防御反应与免疫稳态。若辅助T细胞功能异常,往往会引起免疫相关疾病,如感染不耐受、过敏反应、自身免疫性疾病或肿瘤免疫逃逸。

辅助T细胞的分化

Naive CD4+ T细胞由T细胞受体(TCR)、共刺激分子分化抗原簇CD28同时参与而被活化,同时由表达主要组织相容性复合体(MHCII类的抗原呈递细胞(APC)对naive CD4+ T细胞进行活化,如巨噬细胞、树突状细胞和B细胞。活化信号是通过TCRMHC II类分子表面上的同源抗原肽的识别和结合,并通过多重共刺激信号来传导的。当淋巴组织中的Naive CD4⁺ T细胞获得完全激活后,就会迅速增殖分化为不同的辅助性T细胞(图2),例如Th1Th2Th17细胞

辅助T细胞与疾病

辅助T细胞亚群的平衡与免疫稳态息息相关,其功能失衡往往是疾病发生和发展的根本原因之一。例如,Th1/Th2比例失衡与过敏或感染易感性相关;Th17细胞过度活化与类风湿关节炎、银屑病等自身免疫病密切相关;Treg功能缺陷则可能导致移植物排斥和免疫耐受失败。在肿瘤免疫治疗中,不同辅助T细胞亚群对疗效预测和疗程监控也有重要参考价值。因此,准确、系统地检测辅助T细胞亚群,对科研和临床均具有重要意义。

在科研和临床研究中,流式细胞术是检测辅助T细胞及其亚群的重要手段,通过分析其表面分子、分泌因子及转录因子,可以实现对不同亚群的高效识别与定量。
