生物活性IGN523 elicits strong ADCC activity in Ramos, KG-1 and OCI-AML-3 cells, with EC 50 s of 18.6 ng/mL, 73.8 ng/mL and 83.3 ng/mL, respectively, does not reduce ADCC activity in IGN523 cells [1]. IGN523 (2 ug/mL, 0.5-48 h) elicits Caspase-dependent apoptosis, increases lysosomal membrane permeability and decreases amino acid transport in Ramos, KG-1 and OCI-AML-3 cells [1]. Apoptosis Analysis [1] Cell Line: Ramos, KG-1 and OCI-AML-3 cells Concentration: 2 ug/mL Incubation Time: 48 h Result: Decreased cell viability in the presence of AbXL (10 ug/mL) (Ramos: 70% reduction; OCI-AML-3: 76% reduction; KG-1: 80% reduction), but not when used alone. Increased caspase-3 and caspase-7 activity (Ramos: 3.3-fold; OCI-AML-3: 7.5-fold and KG-1: 6.6-fold). . IGN523 (15 mg/kg, i.p., once a week, twice/three times) inhibits tumor growth in Ramos and Dau xenografts mice model [1]. IGN523 (15 mg/kg, i.p., once a week, twice/three times/four times) inhibits tumor growth in AML and primary NSCLC xenograft mice models [1].

背景

Amino acid transporter heavy chain SLC3A2 (CD98) is a ~57 kDa protein. Acts as a chaperone that facilitates biogenesis and trafficking of functional transporters heterodimers to the plasma membrane. Forms heterodimer with SLC7 family transporters (SLC7A5, SLC7A6, SLC7A7, SLC7A8, SLC7A10 and SLC7A11), a group of amino-acid antiporters. Heterodimers function as amino acids exchangers, the specificity of the substrate depending on the SLC7A subunit. Heterodimers SLC3A2/SLC7A6 or SLC3A2/SLC7A7 mediate the uptake of dibasic amino acids. Heterodimer SLC3A2/SLC7A11 functions as an antiporter by mediating the exchange of extracellular anionic L-cystine and intracellular L-glutamate across the cellular plasma membrane.
